    Vision of breeding for organic agriculture

    This report describes the results of research into the vision of breeding systems within organic agriculture in the Netherlands. The purpose of the research was to arrive at a vision for breeding in organic agriculture by means of interviews and discussions with organic livestock farmers and social organisations. The research was prompted by the fact that, as things stand, organic livestock farmers generally have to rely on the conventional breeding supply. Neither the breeding method not the animal type produced meet the requirements of organic agriculture. Interest in breeding has increased because organic agriculture is expanding, and as yet too little attention has been paid to the development of specific organic breeding programmes and associated legislation. In recent decades conventional agriculture and breeding have tended more and more towards industrialisation and uniformity with breeding becoming a multinational concern. The breeding organisations have no incentive at present to provide special services for organic agriculture as the market is too small and the costs are too high. Taking the current breeding situation as the starting point, a number of scenarios are described for each animal sector which could gradually lead to a system of breeding which is more organic both in its aims and in the chain-based approach. The naturalness of the breeding techniques is an important factor in considering the available options. The different scenarios served as a basis for the interviews and discussions with livestock farmers and social organisations. We have established that the farmers and organisations consider it important for organic agriculture to work on developing a breeding system which follows the principles of organic agriculture. The most important reasons for this are that: (1) consumers expect all production factors in the chain to be of organic origin, (2) most livestock farmers currently use conventional breeding techniques which fall short of the organic requirements on a number of points, such as the use of artificial reproduction techniques and mono-functional breeding for production. For the development of organic breeding, livestock farmers feel that in the first instance the use of artificial reproduction techniques, including cloning and transgenic techniques, should be restricted. Next the livestock farmers and social organisations want breeding to be adapted to or based on the organic environment. There is a suspicion that owing to genotype-environment interaction (G x E) conventionally-bred animals cannot adapt well to the organic environment, and this leads to health and welfare problems. The farmers would like to see this development taking place within 5 to 10 years. It must however proceed one step at a time since the farmers cannot yet form a complete picture of the impact of all the different factors. Most of the people involved see the ideal form of breeding, with natural reproduction and regional or farm-specific selection, as a standard to be achieved in the distant future. At the moment most livestock farmers have neither the knowledge nor the socio-economic means to set up such breeding programmes. The development of breeding and the associated legislation require an international approach, for which suitable contacts must be sought in other countries. The final chapter of this report looks in more detail at the steps to be taken in each sector. Ideally developments should probably be initiated and supervised by a central body, such as an organic breeding foundation, which could be set up to govern the breeding of all farm animals

    It is uncertain whether animals which have been bred for conventional production are capable of optimum performance in organic conditions. In conventional agriculture there is a movement towards maximum control of production conditions in order to optimise animals' yield in intensive production systems. By contrast, organic agriculture is based on natural processes and closed cycles, and takes into account the underlying connections between production factors. Following organic ideology, production capacity should be curtailed by acting in accordance with guiding principles such as naturalness, animal welfare, efficient use of fossil fuels in the farm cycle, and agri-biodiversity (IFOAM, 1994). Organic production should be tied to the land, with farms preferably being self-sufficient mixed farms with closed cycles. An additional point of concern are the reproduction techniques used in conventional breeding. Artificial insemination (AI) and embryo transfer (ET) are commonplace in conventional animal breeding. But these techniques are 'artificial' and they deprive animals of natural mating behaviour and negatively affect the animals' welfare and integrity. By bringing in animals from conventional agriculture, organic farmers are indirectly making use of these techniques. These and other concerns have led to the project 'Organic breeding: a long way to go', which aims to lay down clear visions and an action plan for an organic breeding system

    Coronary calcium mass scores measured by identical 64-slice MDCT scanners are comparable: a cardiac phantom study

    To assess whether absolute mass scores are comparable or differ between identical 64-slice MDCT scanners of the same manufacturer and to compare absolute mass scores to the physical mass and between scan modes using a calcified phantom. A non-moving anthropomorphic phantom with nine calcifications of three sizes and three densities was scanned 30 times on three 64-slice MDCT scanners of manufacturer A and on three 64-slice MDCT scanners of manufacturer B in both sequential and spiral scan mode. The mean mass scores and mass score variabilities of seven calcifications were determined for all scanners; two non-detectable calcifications were omitted. It was analyzed whether identical scanners yielded similar or significantly different mass scores. Furthermore mass scores were compared to the physical mass and mass scores were compared between scan modes. The mass score calibration factor was determined for all scanners. Mass scores obtained on identical scanners were similar for almost all calcifications. Overall, mass score differences between the scanners were small ranging from 1.5 to 3.4% for the total mass scores, and most differences between scanners were observed for high density calcifications. Mass scores were significantly different from the physical mass for almost all calcifications and all scanners. In sequential mode the total physical mass (167.8 mg) was significantly overestimated (+2.3%) for 4 out of 6 scanners. In spiral mode a significant overestimation (+2.5%) was found for system B and a significant underestimation (−1.8%) for two scanners of system A. Mass scores were dependent on the scan mode, for manufacturer A scores were higher in sequential mode and for manufacturer B in spiral mode. For system A using spiral scan mode no differences were found between identical scanners, whereas a few differences were found using sequential mode. For system B the scan mode did not affect the number of different mass scores between identical scanners. Mass scores obtained in the same scan mode are comparable between identical 64-slice CT scanners and identical 64-slice CT scanners on different sites can be used in follow-up studies. Furthermore, for all systems significant differences were found between mass scores and the physical calcium mass; however, the differences were relatively small and consistent

    Genetski i socio-ekonomski ujednačena selekcija proizvodnih (mlijeko i govedina) i finkcionalnih osobina (zdravlje, plodnost,djelotvornost iskorištavanja hrane i mliječnost) u mliječnog goveda traži ispravne ekonomske vrijednosti. lspravne relativne razine ekonomskih vrijednosti osobina daju optimalne razine genetskog poboljšanja u skladu s budućim uvjetima proizvodnje; ispravne apsolutne razine važne su za točno izračunavanje ekonomske dobiti uzgojnih programa. Izvođenje ekonomske vrijednosti zahtijeva dobru ekonomsku osnovu, pravilnu metodologiju u odnosu na modele, ukljudujući fiziološko oblikovanje proizvodnje životinja, ekonomičnosti farme i socijalnih aspekata i odgovarajuće pretpostavke uvjetra buduće proizvodnje. Područje definicije cilja uzgoja zanimljivo je osobito zbog toga što se znanje o oblikovanju poboljšava a nesigurni uvjeti buduće proizvodnje neprestano mijenjaju. Osim toga, iako se istraživanja provode već dugo vremena praktična integracija funkcionalnih osobina u ciljevima uzgoja mliječnog goveda još uvijek je veliki izazov za uzgajivače životinja. Ciljevi izvještaja ove radne grupe su: a) predstaviti definiciju skupine osobina uzgojnih ciljeva, b) raspraviti o metodologiji izvođenja ekonomskih vrijednosti, c) dati sažetak literature o ekonomskim vrijednostima funkcionalnih osobina, d) raspraviti o mogućem opravdanju razlike u definiciji uzgojnih ciljeva nekih zemalja, područja i/ili individualnih farma, i e) raspraviti o mogućim budućim tendencijama što bi mogle utjecati na uzgojne ciljeve promjenom ekonomskih vrijednosti.A genetically and socio-economically balanced selection on production (milk and beef) and functional traits (health, fertility, efficiency of feed utilisation and milkability) in dairy cattle requires correct economic values. Correct relative levels of economic values of traits give optimum levels of genetic improvement according to future production circumstances; correct absolute levels are important for an accurate calculation of economic revenues of breeding programmes. The derivation of economic values requires a good theoretical basis, proper methodology in terms of models including physiological modelling of animal production, farm economics and social aspects, and appropriate assumptions on future production circumstances. The field of breeding goal definition is of ongoing interest, especially because knowledge on modelling is improving and on uncertain future production circumstances are continuously changing. Moreover, although research has been undertaken for a long period, the practical integration of functional traits in dairy cattle breeding goals is still a major challenge for animal breeders. The aims of this working group report are: a. to present the definition of a set of breeding goal traits, b. to discuss the methodology in deriving economic values, c. to present a summary of literature on economic values of functional traits, d. to discuss a possible justification of differences in breeding goal definition for countries, regions, and/or individual farms, and e. to discuss possible future trends potentially influencing breeding goals by changing economic values

    Mitotic spindles are self-organizing protein machines that harness teams of multiple force generators to drive chromosome segregation. Kinesins are key members of these force-generating teams. Different kinesins walk directionally along dynamic microtubules, anchor, crosslink, align and sort microtubules into polarized bundles, and influence microtubule dynamics by interacting with microtubule tips. The mechanochemical mechanisms of these kinesins are specialized to enable each type to make a specific contribution to spindle self-organization and chromosome segregation

    Sorafenib in patients with advanced biliary tract carcinoma: a phase II trial

    BACKGROUND: Advanced biliary tract carcinoma has a very poor prognosis, with chemotherapy being the mainstay of treatment. Sorafenib, a multikinase inhibitor of VEGFR-2/-3, PDGFR-beta, B-Raf, and C-Raf, has shown to be active in preclinical models of cholangiocarcinoma. METHODS: We conducted a phase II trial of single-agent sorafenib in patients with advanced biliary tract carcinoma. Sorafenib was administered at a dose of 400 mg twice a day. The primary end point was the disease control rate at 12 weeks. RESULTS: A total of 46 patients were treated. In all, 26 (56%) had received chemotherapy earlier, and 36 patients completed at least 45 days of treatment. In intention-to-treat analysis, the objective response was 2% and the disease control rate at 12 weeks was 32.6%. Progression-free survival (PFS) was 2.3 months (range: 0-12 months), and the median overall survival was 4.4 months (range: 0-22 months). Performance status was significantly related to PFS: median PFS values for ECOG 0 and 1 were 5.7 and 2.1 months, respectively (P=0.0002). The most common toxicities were skin rash (35%) and fatigue (33%), requiring a dose reduction in 22% of patients. CONCLUSIONS: Sorafenib as a single agent has a low activity in cholangiocarcinoma. Patients having a good performance status have a better PFS. The toxicity profile is manageable

    Concurrent chemoradiotherapy with low dose weekly gemcitabine in stage III non-small cell lung cancer

    BACKGROUND: Combined chemoradiotherapy (CRT) is the treatment of choice for stage III NSCLC. Gemcitabine (G) is a novel deoxycitidine analogue that has been proven to be a potent radiosensitizer. Twenty-two consecutive patients were treated with concurrent CRT to demonstrate the tolerability and efficacy of low dose G given weekly as radiosensitizer in stage III NSCLC. METHODS: Patients with KPS ≥70, adequate bone marrow reserve, with no prior radiotherapy (RT) and surgery were included. Eighteen patients had received prior induction chemotherapy (CT). G (75 mg/m(2)/week) was infused over 1 hour for 6 weeks. Thoracic RT was given two hours later over 6 weeks at 1.8 Gy/day fractions (total dose of 61.2 Gy). Pulmonary toxicity was evaluated with computed tomography scans in 6 weeks. RESULTS: Median age was 60 years (range, 48–75), median follow-up was 15 months (range, 2–40). Sixty-eight percent of patients were male and median KPS score was 90. Conformal 3D-RT planning was used in 64% of patients. G was given for a median of 5 weeks (range 1–9). Twelve patients (54.6%) received all planned CT. G was stopped because of intolerance in 6 and death in 2 patients. Seven patients (31.8%) had radiation pneumonitis. Twenty patients were evaluated for overall response, 1 patient (4.5%) had clinical CR, 81.8% had PR while 9.5% had SD. Median overall survival (OS) was 14 ± 5 months (95% CI 3–25). One- and 2-year OS rates were 55% and 38%. Sixteen patients died of disease-related events (6 with progression of primary tumor, 8 due to metastatic disease), 2 patients died of other causes. One- and 2-year progression-free survival and local control rates were 56%, 27% and 79%, 51%, respectively. CONCLUSION: G might be used as radiosensitizer for patients with stage III NSCLC who could not receive full doses CT with concurrent RT

    Weekly gemcitabine plus 24-h infusion of high-dose 5-fluorouracil/leucovorin for locally advanced or metastatic carcinoma of the biliary tract

    [[abstract]]Both gemcitabine and weekly 24-h infusion of high-dose 5-fluorouracil/leucovorin (HDFL) have shown promising antitumour activity for patients with locally advanced or metastatic carcinoma of the biliary tract (CBT). From April 1999 through December 2002, 30 patients with inoperable CBT were treated with gemcitabine 800 mg m(-2), intravenous infusion for 30 min, followed by 5-FU, 2000 mg m(-2) and leucovorin, 300 mg m(-2), intravenous infusion for 24 h, on day 1, 8 and 15, every 4 weeks. A total of 166 cycles were given (median of four cycles per patient, range 1-24 cycles). Response was evaluable in 28 patients and toxicity in 29 patients. Partial response was obtained in six patients, stable disease in 13, while progressive disease occurred in nine. The objective response rate was 21.4% (95% CI: 5.2-37.6%). The most common grade 3 or 4 toxicity was infection (nine patients). Other types of grade 3 or 4 toxicity included leucopenia (four patients), thrombocytopenia (three patients), anaemia (three patients), nausea/vomiting (two patients) and elevation of liver transaminases (three patients). As of 30 September 2003, the median progression-free survival was 3.7 months (95% CI: 2.8-4.6 months) and the median overall survival was 4.7 months (95% CI: 0.8-8.6 months). Our data suggest that weekly gemcitabine plus HDFL is modestly active with acceptable treatment-related toxicity for patients with advanced CBT

    Genetic loci associated with chronic obstructive pulmonary disease overlap with loci for lung function and pulmonary fibrosis.

    Chronic obstructive pulmonary disease (COPD) is a leading cause of mortality worldwide. We performed a genetic association study in 15,256 cases and 47,936 controls, with replication of select top results (P < 5 × 10(-6)) in 9,498 cases and 9,748 controls. In the combined meta-analysis, we identified 22 loci associated at genome-wide significance, including 13 new associations with COPD. Nine of these 13 loci have been associated with lung function in general population samples, while 4 (EEFSEC, DSP, MTCL1, and SFTPD) are new. We noted two loci shared with pulmonary fibrosis (FAM13A and DSP) but that had opposite risk alleles for COPD. None of our loci overlapped with genome-wide associations for asthma, although one locus has been implicated in joint susceptibility to asthma and obesity. We also identified genetic correlation between COPD and asthma. Our findings highlight new loci associated with COPD, demonstrate the importance of specific loci associated with lung function to COPD, and identify potential regions of genetic overlap between COPD and other respiratory diseases
